89Zr-AMA is an immuno-PET imaging agent consisting of a monoclonal antibody (AMA) specific for mesothelin, radiolabeled with the positron-emitting isotope zirconium-89 (89Zr). Mesothelin is a cell-surface glycoprotein that is highly overexpressed in several epithelial malignancies, including pancreatic adenocarcinoma, mesothelioma, and ovarian cancer, while showing restricted expression in normal tissues. By binding specifically to mesothelin, 89Zr-AMA allows for the non-invasive visualization and quantification of antigen expression and tracer biodistribution within tumors using Positron Emission Tomography (PET). This radiopharmaceutical is primarily utilized in clinical and preclinical research to evaluate the tumor-targeting efficiency of mesothelin-directed therapies, such as antibody-drug conjugates (ADCs), and to assist in patient stratification by confirming the presence and accessibility of the target antigen.
