Drug intelligence / Profile preview
89Zr-DFO-PEG-D3 is a radiolabeled nanobody-based positron emission tomography (PET) imaging agent developed by researchers at Boston Children's Hospital and Harvard Medical School. It consists of the idiotype-specific nanobody clone D3, which is site-specifically sortase-conjugated to the chelator deferoxamine-polyethylene glycol (DFO-PEG) and labeled with the positron-emitting radioisotope zirconium-89 (89Zr). The nanobody D3 specifically targets the 2D2 T-cell receptor (TCR) idiotype, which is expressed on myelin oligodendrocyte glycoprotein (MOG)-specific CD4+ T cells. Autoreactive MOG-specific T cells are key drivers of neuroinflammation and demyelination in multiple sclerosis (MS). By binding to the 2D2 TCR, 89Zr-DFO-PEG-D3 enables non-invasive, high-contrast, longitudinal PET imaging of antigen-specific T cells in vivo, allowing researchers to monitor T-cell homing to secondary lymphoid organs and infiltration into the central nervous system in experimental autoimmune encephalomyelitis (EAE) models.
