89Zr-DFOMalDAR2SC16-MB1 is an immuno-PET (positron emission tomography) imaging agent designed to target Delta-like protein 3 (DLL3). It consists of the SC16 monoclonal antibody, a humanized IgG1 that binds with high affinity to DLL3, conjugated to the desferrioxamine (DFO) chelator via a maleimide (Mal) linker with a drug-to-antibody ratio (DAR) of 2, and radiolabeled with the positron-emitting isotope Zirconium-89 (89Zr). DLL3 is an atypical Notch ligand that is significantly upregulated on the surface of small cell lung cancer (SCLC) and other high-grade neuroendocrine tumors, while expression in normal tissues is minimal. This tracer facilitates the non-invasive assessment of DLL3 expression, aiding in patient stratification for DLL3-targeted treatments (such as antibody-drug conjugates or T-cell engagers) and monitoring therapeutic efficacy. The "MB1" designation refers to a specific site-specific conjugation methodology or variant used to ensure a homogenous DAR and optimal pharmacokinetic properties.
