89Zr-KN035 is a radiolabeled immuno-PET imaging agent composed of the anti-PD-L1 domain antibody KN035 conjugated to the chelator DFO and labeled with zirconium-89 (89Zr).** KN035 is a recombinant humanized single-domain antibody Fc fusion protein that specifically targets programmed death-ligand 1 (PD-L1), an immune checkpoint molecule expressed on tumor cells and various immune cells. By binding PD-L1, KN035 blocks its interaction with PD-1, thereby potentially enhancing T-cell mediated antitumor immunity. The radiolabeling with zirconium-89 enables noninvasive positron emission tomography (PET) imaging to visualize and quantify PD-L1 expression in vivo. This allows for sensitive detection of primary and metastatic tumors expressing PD-L1, monitoring of therapeutic efficacy in patients undergoing immunotherapy, assessment of biodistribution, and prediction of patient prognosis. The agent has demonstrated high tumor-to-background ratios in both preclinical models and clinical studies involving patients with advanced solid tumors[1][4][5][6][8][9].
