8B4 VHH CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy targeting CD70, a protein highly expressed in various hematopoietic and solid tumors, including renal cell carcinoma (RCC), but absent in normal tissues. Developed by HRAIN Biotechnology, this therapy utilizes a variable heavy chain-only (VHH) antibody fragment (nanobody) derived from an immunized alpaca as the antigen-binding domain. In preclinical studies presented at ASCO 2023, the 8B4 clone was one of several VHH-based CAR-T constructs evaluated for its ability to eliminate CD70-positive tumor cells. While clones like 11C9 and 2A5 showed superior expansion and persistence in re-challenge models, 8B4 demonstrated effective in vitro cytotoxicity against CD70-expressing targets.
