8F3 is a human monoclonal antibody designed to target Pancreatic Adenocarcinoma Up-regulated Factor (PAUF), a soluble protein that is significantly overexpressed in pancreatic cancer and plays a pivotal role in tumor progression and metastasis. By binding to PAUF, 8F3 neutralizes its activity, leading to the inhibition of cancer cell migration, invasion, adhesion, and proliferation. Preclinical studies have demonstrated that 8F3 treatment downregulates the expression of CXCR4, a receptor critical for organ-specific metastasis, and alters intracellular signaling pathways induced by PAUF. Developed by the Korea Research Institute of Bioscience and Biotechnology (KRIBB), 8F3 has shown efficacy in orthotopic xenograft mouse models, both as a monotherapy and in additive combination with gemcitabine, resulting in tumor growth retardation and suppression of metastatic spread.
