8F4 CAR-modified cord blood iNKT cells are **allogeneic cell therapy products** consisting of invariant natural killer T (iNKT) cells isolated from human umbilical cord blood and genetically engineered to express a chimeric antigen receptor (CAR) known as 8F4. The 8F4 CAR targets the PR1 peptide presented by HLA-A2, an antigen associated predominantly with acute myeloid leukemia (AML). These cells combine the intrinsic anti-tumor and immunomodulatory activity of iNKT cells with the antigen-specific cytotoxicity conferred by the 8F4 CAR. Compared to adult-derived iNKT cells, cord blood-derived 8F4 CAR-iNKT cells exhibit higher expansion capability, a purer and consistently CD4-positive phenotype, increased CD62L (central memory marker) expression, and a Th2/Th10-biased cytokine production, which may enhance safety by reducing the risk of severe cytokine release syndrome. In preclinical models, these cells display potent cytotoxicity against AML in vitro and in vivo, with superior persistence and expansion potential relative to adult counterparts[1][2].
