8H8-SDIE is an Fc-optimized monoclonal antibody targeting CD276 (B7-H3), an immune checkpoint protein overexpressed in various solid and hematological malignancies. Developed by the University of Tübingen, the antibody is an engineered variant of the 8H8 clone, incorporating S239D and I332E (SDIE) mutations in the Fc region to significantly enhance its binding affinity to the CD16 (FcγRIIIa) receptor on natural killer (NK) cells. This modification promotes potent antibody-dependent cellular cytotoxicity (ADCC) against tumor cells. The drug is currently in preclinical development for indications including colorectal cancer, non-small cell lung cancer, ovarian cancer, and acute myeloid leukemia.
