8Qm-polyleucine conjugate is a therapeutic peptide-based vaccine candidate developed for the treatment of Human Papillomavirus (HPV)-related malignancies, particularly cervical cancer. The vaccine consists of a specific peptide epitope derived from the HPV-16 E7 protein, known as 8Qm, which is covalently conjugated to a polyleucine (poly-L-leucine) tail. This polyleucine component serves as a self-adjuvating delivery system, facilitating the self-assembly of the conjugate into nanoparticles or its incorporation into liposomal formulations. By utilizing the hydrophobic nature of the polyleucine tail, the vaccine stimulates robust cellular immune responses, including the activation of dendritic cells and the induction of cytotoxic T-lymphocytes, without the requirement for traditional external adjuvants. Preclinical studies have demonstrated that liposomal formulations of this conjugate can significantly inhibit tumor growth and eradicate established HPV-16-positive tumors in mouse models.
