8R-70CAR T-cells are an investigational chimeric antigen receptor (CAR) T-cell therapy designed to target the CD70 protein, which is overexpressed in various hematologic and solid tumors. These T-cells are uniquely modified to constitutively express CXCR2 (the IL-8 receptor), a modification intended to enhance the cells' ability to migrate toward IL-8-rich tumor microenvironments, such as those found in glioblastoma and acute myeloid leukemia (AML). This "8R" modification aims to overcome common CAR-T limitations like poor trafficking and fitness by harnessing cytokine gradients. The therapy is currently being evaluated in Phase I clinical trials for glioblastoma and is undergoing preclinical investigation for AML, where it may be used in combination with epigenetic modifiers like azacitidine to increase target density.
