9-(4-hydroxybutyl)-N2-phenylguanine (HBPG) is a synthetic small molecule belonging to the hypoxanthines, a class of purine derivatives. It acts as a potent inhibitor of viral thymidine kinase, particularly from herpes simplex virus (HSV) types 1 and 2. Unlike traditional antiviral nucleoside analogs, HBPG does not exhibit direct antiviral activity but instead blocks viral reactivation from latency by inhibiting the phosphorylation activity of viral thymidine kinase. Preclinical studies have shown that HBPG can suppress herpesvirus reactivation in animal models and reduce recurrences of herpetic keratitis[1][4][5][6]. The compound has been investigated experimentally but has not reached clinical approval or advanced clinical development stages.
