Overview

**9-deazainosine** is a synthetic purine nucleoside analog belonging to the class of c-glycosyl compounds and pyrrolopyrimidines[1][2]. It acts as an inosine analog and has demonstrated inhibitory activity against various pathogens by interfering with purine metabolism. Mechanistically, it inhibits the enzyme purine nucleoside phosphorylase (PNP), which plays a key role in the purine salvage pathway[5]. This inhibition disrupts nucleotide synthesis in certain organisms. Preclinical studies have shown that 9-deazainosine is effective against Pneumocystis carinii pneumonia in immunocompromised rats[7] and exhibits potent activity against Trypanosoma brucei (the causative agent of African trypanosomiasis), especially when used in combination with eflornithine (DFMO)[10]. The compound has been studied primarily as an experimental therapeutic agent for infectious diseases.
