The 9-peptide breast cancer vaccine is an investigational peptide-based immunotherapy developed by Craig L. Slingluff Jr. and colleagues at the University of Virginia. It is designed to induce a multi-antigenic T-cell response against micrometastatic disease in patients with stage I-IV breast cancer. The vaccine consists of nine MHC class I-restricted peptides derived from six tumor-associated antigens: MAGE-A1, MAGE-A3, MAGE-A10, carcinoembryonic antigen (CEA), NY-ESO-1, and HER2. To enhance immunogenicity, the vaccine is administered in combination with poly-ICLC, a Toll-like receptor 3 (TLR3) agonist that serves as an adjuvant, and a helper peptide derived from tetanus toxoid to provide CD4+ T-cell support.
