**90Y-anti-ROBO1** is a radiolabeled monoclonal antibody that targets the ROBO1 membrane protein, which is implicated in tumor metastasis and angiogenesis. The antibody (clone B5209B) is conjugated with DOTA and subsequently radiolabeled with yttrium-90 (^90Y), resulting in a radiopharmaceutical used for radioimmunotherapy (RIT). Its mechanism involves binding specifically to ROBO1-expressing tumor cells, delivering cytotoxic radiation via ^90Y, causing tumor growth suppression, induction of apoptosis, and reduction of proliferative index in preclinical models. It has demonstrated antitumor effects in xenograft models of hepatocellular carcinoma and small cell lung cancer by inducing cell degeneration and necrosis at tumor sites. Systemic toxicity is mainly transient bone marrow suppression. Primary indications being studied are hepatocellular carcinoma and small cell lung cancer, in ROBO1-positive tumors[1][2][3].
