90Y-BU-12 is a radioimmunotherapy (RIT) agent consisting of the murine monoclonal antibody BU-12, which specifically targets the CD19 antigen, labeled with the radioisotope Yttrium-90 (90Y). CD19 is a transmembrane glycoprotein expressed on the surface of B-lineage cells from the early pro-B cell stage until terminal differentiation into plasma cells, making it a highly specific target for B-cell malignancies. The mechanism of action involves the antibody-mediated delivery of high-energy beta radiation from the 90Y isotope directly to CD19-positive tumor cells. This localized radiation induces lethal DNA damage and apoptosis in the targeted cells and provides a "bystander effect," where the emitted radiation can kill adjacent tumor cells within the radiation range, even if they lack the target antigen. 90Y-BU-12 has been primarily investigated for the treatment of refractory B-cell non-Hodgkin lymphoma and acute lymphoblastic leukemia.
