90Y-DOTA-30F11 is a radiopharmaceutical conjugate designed for radioimmunotherapy (RIT) of hematologic malignancies, specifically acute myeloid leukemia (AML). It consists of the rat monoclonal antibody 30F11, which targets the murine CD45 antigen (leukocyte common antigen), conjugated via the DOTA chelator to the beta-emitting radionuclide Yttrium-90 (90Y). CD45 is highly expressed on the surface of most nucleated hematopoietic cells and leukemic blasts, making it an ideal target for delivering radiation to the bone marrow and spleen while sparing non-hematologic organs. In preclinical murine models, 90Y-DOTA-30F11 has demonstrated superior therapeutic efficacy compared to 177Lu-labeled counterparts, providing dose-dependent survival benefits with transient myelotoxicity. The drug is primarily developed and studied by researchers at the Fred Hutchinson Cancer Research Center as part of a broader effort to optimize RIT conditioning regimens for hematopoietic cell transplantation.
