90Y-DOTA-cG250 is a radiopharmaceutical conjugate consisting of the chimeric monoclonal antibody girentuximab (cG250) linked to the beta-emitting radioisotope yttrium-90 (90Y) through the chelator DOTA. The antibody component specifically targets carbonic anhydrase IX (CAIX), an enzyme that is highly overexpressed on the cell surface of clear cell renal cell carcinomas (ccRCC) but has very limited expression in healthy tissues. Upon binding to the CAIX antigen on tumor cells, the yttrium-90 delivers high-energy beta radiation locally, causing DNA damage and cell death. This targeted radioimmunotherapy approach is designed to treat advanced or metastatic renal cell carcinoma while minimizing systemic toxicity to non-target organs.
