90Y-DOTATATE is a radiolabeled peptide used in peptide receptor radionuclide therapy (PRRT) for the treatment of neuroendocrine tumors (NETs). It consists of the somatostatin analogue Tyr3-octreotate conjugated to the chelator DOTA and labeled with the beta-emitting radioisotope yttrium-90. The drug binds with high affinity to somatostatin receptor subtype 2 (SSTR2), which is overexpressed on many NET cells. Upon binding, it delivers targeted radiation to tumor cells, leading to DNA damage and cell death while sparing most normal tissues. Its longer beta range compared to other isotopes makes it particularly effective for treating larger tumors[1][5]. Clinical studies have shown disease stabilization or partial remission in a significant proportion of patients with advanced or metastatic NETs[5][2].
