90Y-epratuzumab tetraxetan is a radioimmunotherapeutic agent composed of the humanized monoclonal antibody epratuzumab, which targets the CD22 antigen on B lymphocytes, conjugated to the radioactive isotope yttrium-90 via a chelator (tetraxetan/DOTA). The drug delivers targeted beta radiation to CD22-expressing tumor cells, resulting in cytotoxicity primarily through DNA damage. It was developed for use in B-cell malignancies such as non-Hodgkin lymphoma (NHL) and acute lymphoblastic leukemia (ALL), with clinical studies showing durable responses especially when administered in repeated low doses or combined with other therapies like rituximab or veltuzumab. Despite promising early results, further development was discontinued due to loss of industrial support and it has not received regulatory approval[1][3][5][6][7][9].
