90Y-huA33 is a radioimmunotherapeutic agent consisting of the humanized monoclonal antibody huA33 conjugated to the high-energy beta-emitting radioisotope Yttrium-90 (90Y) via the bifunctional chelator CHX-A″-DTPA. The antibody component, huA33, specifically targets Glycoprotein A33 (GPA33), a transmembrane protein that is highly and homogeneously expressed in over 95% of primary and metastatic colorectal cancers, while having restricted expression in normal intestinal mucosa. By binding to GPA33-positive tumor cells, 90Y-huA33 delivers localized ionizing radiation, which induces DNA damage and apoptosis in the targeted cells and neighboring cells through the crossfire effect. Developed by the Ludwig Institute for Cancer Research, 90Y-huA33 has been investigated in clinical trials for the treatment of colorectal cancer and is frequently used as an isotype control in preclinical studies for other IgG1-based radioimmunotherapies.
