917T BCMA CAR-T is an experimental monovalent chimeric antigen receptor (CAR) T-cell therapy designed to target the B-cell maturation antigen (BCMA), which is highly expressed in multiple myeloma. Developed by researchers at Fujian Medical University Union Hospital, this construct was created as part of a comparative study to evaluate the efficacy of monovalent versus bivalent CAR designs, using the structural framework of ciltacabtagene autoleucel (Cilta-cel) as a reference. In preclinical models, 917T demonstrated cytotoxic activity against multiple myeloma cell lines, although it showed lower killing efficacy compared to the bivalent 353/917T construct. Research indicates that its therapeutic effect may be enhanced by inhibiting the NF-κB pathway, which is activated in tumor cells during co-culture with these CAR-T cells.
