Overview

**99mTc-blue-biotin-liposomes** is a preclinical liposomal radiopharmaceutical imaging agent developed for sentinel lymph node radiolocalization. The formulation contains biotin-coated liposomes that co-encapsulate patent blue dye and glutathione, with technetium-99m incorporated through an HMPAO-glutathione radiolabeling method. After local administration, separately administered avidin binds the biotin surface, causing liposome aggregation and retention in the first draining lymph node; the patent blue dye enables visual identification and the technetium-99m label enables scintigraphic localization. It has also been investigated in rodents for avidin-mediated targeting of liposomes to peritoneal, mediastinal, and diaphragmatic lymph nodes.
