99mTc-HYNIC-cHK is a technetium-99m labeled radiopharmaceutical designed for noninvasive imaging of integrin αvβ6 expression using single-photon emission computed tomography (SPECT). The tracer consists of a disulfide-cyclized peptide (cHK) conjugated to the bifunctional chelator 6-hydrazinonicotinic acid (HYNIC) and labeled with 99mTc. Integrin αvβ6 is a cell surface receptor that is significantly upregulated in various cancers, particularly pancreatic cancer, and in fibrotic conditions like idiopathic pulmonary fibrosis (IPF), while remaining nearly undetectable in normal tissues. Compared to its linear counterpart (99mTc-HYNIC-HK), the cyclic version demonstrates improved metabolic stability and higher tumor-to-muscle ratios, providing high-contrast images for disease diagnosis and therapy monitoring.
