99mTc-NM-02 is a technetium-99m-labeled single-domain antibody (nanobody) that specifically targets human epidermal growth factor receptor 2 (HER2). It is designed as a molecular imaging agent for noninvasive detection of HER2 expression in cancer patients, primarily using SPECT/CT imaging. The drug binds with high affinity and specificity to HER2, allowing visualization of HER2-positive tumors and metastases. This enables assessment of HER2 status without the need for repeated biopsies and can reveal intra-tumoral heterogeneity. Clinical studies have shown that 99mTc-NM-02 has favorable biodistribution (mainly accumulating in kidneys and liver), reasonable radiation doses, no significant adverse effects, and effective tumor targeting. It is being developed as a diagnostic tool to guide HER2-targeted therapy decisions in breast cancer[1][5][7].
