Drug intelligence / Profile preview
99mTc-UBI29-41-Cy5 is a bacteria-specific hybrid (dual-labeled) imaging tracer developed by the Leiden University Medical Center (LUMC) for the real-time visualization and quantification of bacterial infections, particularly periprosthetic joint infections (PJI) on orthopedic prostheses. The tracer consists of the cationic antimicrobial peptide fragment ubiquicidin 29-41 (UBI29-41), which is conjugated to both the radioactive isotope technetium-99m (99mTc) for single-photon emission computed tomography (SPECT) or gamma imaging, and the near-infrared fluorescent dye Cyanine 5 (Cy5) for optical/fluorescence imaging. UBI29-41 targets the anionic components of bacterial cell membranes, such as lipopolysaccharide (LPS) in Gram-negative bacteria and lipoteichoic acid (LTA) in Gram-positive bacteria, through electrostatic interactions. This hybrid design allows for qualitative real-time fluorescence-guided surgery (e.g., during debridement, antibiotics, and implant retention [DAIR] procedures) to be combined with quantitative nuclear imaging to monitor the efficacy of cleaning strategies.
