Overview

9A8-41BBζ is a chimeric antigen receptor (CAR) T cell therapy specifically engineered to target CD22, a surface antigen commonly expressed in B cell malignancies such as B-cell acute lymphoblastic leukemia (B-ALL). The construct comprises the 9A8 single-chain variable fragment (scFv) recognizing CD22, a 4-1BB (CD137) costimulatory domain, and a CD3 zeta (ζ) signaling domain. In preclinical studies and early clinical assessment, 9A8-41BBζ exhibits potent cytotoxicity against CD22-positive targets, including those with low antigen density, and improves IL-2 secretion and proliferation of modified T cells. It is being evaluated to address antigen escape (relapse with loss of CD19 or low CD22 expression) and is used both alone and in combination (co-transduction) with CAT-41BBζ (CD19 CAR) in children with relapsed/refractory B-ALL[1].
