9D8 VHH CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy developed by HRAIN Biotechnology for the treatment of CD70-positive malignancies, specifically renal cell carcinoma (RCC). The therapy employs a variable heavy chain-only (VHH) antibody fragment, or nanobody, as the antigen-binding domain. This VHH was selected from an alpaca-immunized library for its specificity to CD70, a member of the tumor necrosis factor superfamily that is highly expressed in various solid and hematopoietic tumors but has limited expression in healthy tissues. Preclinical data presented at ASCO 2023 demonstrated that 9D8 VHH CAR-T cells effectively eliminate CD70-positive tumor cells in vitro and exhibit significant anti-tumor activity in xenograft mouse models.
