9E,11E-conjugated linoleic acid (9E,11E-CLA), also known as isolinoleic acid or Mangold's acid, is a specific geometric isomer of linoleic acid with two contiguous double bonds in the all-trans configuration at positions 9 and 11. It is a naturally occurring polyunsaturated fatty acid found in trace amounts in beef fat and dairy products. Unlike more common CLA isomers (such as 9-cis,11-trans), the 9E,11E isomer has demonstrated unique and potent anti-inflammatory, anti-carcinogenic, and anti-atherogenic properties in preclinical research. Its mechanism of action primarily involves the induction of the endogenous anti-inflammatory factor interleukin-1 receptor antagonist (IL-1Ra) through the activation of the phosphoinositide 3-kinase (PI3K) and mechanistic target of rapamycin (mTOR) signaling pathways. This activity leads to the downregulation of several atherogenesis-related genes, including adhesion molecules like ICAM-1 and VCAM-1, and chemokines such as MCP-1. While it shows significant biological potential, 9E,11E-CLA is currently utilized primarily as a research chemical and is not in formal pharmaceutical development by any identified commercial entity.
