9MW2921 is a next-generation antibody-drug conjugate (ADC) developed by Mabwell (Shanghai) Bioscience for the treatment of solid tumors. It utilizes Mabwell's proprietary IDDC (Interchain-Disulfide Drug Conjugate) platform and consists of an innovative antibody molecule linked to a novel topoisomerase I inhibitor payload (TOP1i) via a unique linker. The drug specifically binds to Trop-2 antigen on the tumor cell membrane—leading to internalization, lysosomal trafficking, release of the cytotoxic payload, and subsequent induction of apoptosis in tumor cells. Preclinical studies have demonstrated favorable safety and pharmacokinetic profiles, with controlled on-target and off-target toxicities. As of late 2023/early 2024, it is in phase I/II clinical trials for advanced or metastatic solid tumors.
