Aβ-CoreS is an experimental RNA-based vaccine designed to encode the Aβ1-11 fragment of the amyloid-beta protein. Developed by researchers at the National Institute on Aging (NIA), it is used as an immunization tool to expand antigen-specific CD8+ T cells. In preclinical studies using 5xFAD mouse models of Alzheimer's disease, Aβ-CoreS was found to exacerbate disease symptoms by promoting the infiltration of cytotoxic and exhausted CD8+ T cells into the brain, which in turn increases Aβ plaque accumulation and disease-associated microglia. This research highlights the potential role of adaptive immunity, specifically CD8+ T cells, in the progression of neurodegeneration.
