Aβ1-10-S8R epitope is a peptide-based B-cell epitope vaccine candidate designed for the treatment of Alzheimer's disease (AD). It consists of the first ten amino acids of the amyloid-beta (Aβ) peptide with a specific mutation at the eighth position, where Serine is replaced by Arginine (S8R). To overcome the low immunogenicity of short peptides, the Aβ1-10-S8R epitope is typically conjugated to carrier proteins such as ovalbumin (OVA) or keyhole limpet hemocyanin (KLH). The vaccine aims to induce a robust humoral immune response, specifically increasing immunoglobulin G (IgG) levels that target Aβ aggregates. Preclinical studies in mouse models have demonstrated that this immunization strategy effectively suppresses the expression of amyloid-beta, amyloid-beta precursor protein (APP), and beta-secretase 1 (BACE-1), while simultaneously reducing neuroinflammation by decreasing inflammatory cytokine production from astrocytes and microglia. Furthermore, the vaccine has shown potential in preserving synaptic integrity by increasing levels of key synaptic markers such as synaptophysin, SNAP-23, and PSD-95.
