A-005 is a potential first-in-class, brain-penetrant, allosteric tyrosine-protein kinase 2 (TYK2) inhibitor developed by Alumis for the treatment of neuroinflammatory and neurodegenerative diseases. It acts by inhibiting TYK2, a key enzyme in the JAK/STAT signaling pathway involved in immune cell activation and inflammation. Preclinical studies have shown that A-005 can inhibit tumor growth and prevent osteosarcoma-associated osteolysis via modulation of the PI3K/AKT/mTOR pathway. In clinical trials, it demonstrated central nervous system (CNS) penetration with significant exposure in cerebrospinal fluid and prolonged inhibition of TYK2 activity as measured by phosphorylated STAT proteins. The drug is being developed primarily for multiple sclerosis but also has potential applications in Parkinson's disease and other neuroinflammatory conditions[1][2][4][5].
