Overview

A-1210477 is a potent, cell-permeable, and highly selective small-molecule inhibitor of the anti-apoptotic protein MCL-1. Developed by AbbVie, it functions as a BH3 mimetic by binding with high affinity to the BH3-binding groove of MCL-1, thereby disrupting its interaction with pro-apoptotic proteins such as BIM and NOXA. This displacement facilitates the activation of BAX and BAK, leading to mitochondrial outer membrane permeabilization and the induction of the intrinsic apoptotic pathway. While primarily utilized as a chemical probe in preclinical research, A-1210477 has demonstrated significant utility in sensitizing various cancer types—including melanoma and hematological malignancies—to other BCL-2 family inhibitors like ABT-263 (navitoclax) or ABT-199 (venetoclax), effectively overcoming MCL-1-mediated resistance.
