A-784168 is a potent, orally active small molecule antagonist of the transient receptor potential vanilloid 1 (TRPV1) receptor, originally developed by Abbott Laboratories. It competitively inhibits TRPV1 activation with an IC50 of 25 nM against capsaicin-induced activation of the human receptor. The compound demonstrates significant central nervous system (CNS) penetration and is highly selective for TRPV1 over a panel of 74 other neurotransmitter receptors and ion channels, though it exhibits some secondary activity at TRPM8. In preclinical models, A-784168 has shown efficacy in reducing capsaicin-induced pain, thermal hyperalgesia, and mechanical hypersensitivity, suggesting potential utility in treating acute and chronic inflammatory pain conditions.
