A-dmDT390-bisFv(UCHT1) + pembrolizumab is an investigational combination therapy comprised of a recombinant fusion toxin protein (A-dmDT390-bisFv(UCHT1)) and a monoclonal antibody immune checkpoint inhibitor (pembrolizumab). A-dmDT390-bisFv(UCHT1) is a fusion protein composed of a truncated diphtheria toxin (DT390) linked to two single-chain variable fragments (bisFv) derived from the anti-CD3 monoclonal antibody UCHT1. It targets CD3 on T cells, delivering the toxin to induce cell death. Pembrolizumab is an anti-PD-1 monoclonal antibody that blocks programmed death receptor-1, releasing inhibition of T-cell–mediated immune responses against tumors. The combination is being studied for synergistic anti-tumor activity by promoting T cell killing and releasing immune checkpoint inhibition. The primary indication under investigation is oncology, particularly advanced solid tumors and lymphoid malignancies.
