A07D is a first-in-class, oral peptide drug candidate developed by Endevica Bio that acts as an agonist of the melanocortin 4 receptor (MC4R). It is designed for the treatment of obesity and related metabolic disorders, such as diabetes. By modulating the central melanocortin system, A07D induces weight loss while uniquely promoting the retention of lean muscle mass, addressing a significant limitation of current GLP-1 receptor agonist therapies. Preclinical studies in diet-induced obesity (DIO) rat models have demonstrated that A07D provides durable weight loss that persists even after the cessation of treatment, suggesting it may overcome the weight loss plateaus and rapid weight rebound often seen with existing metabolic treatments. A07D is considered a sister molecule to TCMCB07, which is being developed by the same company for cancer-related cachexia.
