Overview

A10 ARTEMIS T cells are engineered T cells designed to target glioblastoma (GBM) and other EGFR-driven cancers. This cell therapy utilizes the proprietary ARTEMIS® Cell Receptor Platform, developed by Eureka Therapeutics, Inc., which introduces two separate receptors into T cells to provide activation and co-stimulatory signals. Specifically, the A10 antigen-binding fragment (Fab) is fused to the N-terminus of γδTCR-signaling domains (A10-AbTCR), while the A10 single chain variable fragment (scFv) is fused to a costimulatory signaling receptor (A10-CSR). These engineered T cells are cytotoxic to tumor cell lines displaying amplified or overexpressed Epidermal Growth Factor Receptor (EGFR) and its variant EGFRvIII. Preclinical studies in glioblastoma PDX models have shown that A10 ARTEMIS T cells can prolong survival and achieve complete tumor regression in EGFR-amplified cancers.
