A24110He, also known as Lipisense, is a GalNAc-conjugated antisense oligonucleotide (ASO) developed by Lipigon Pharmaceuticals for the treatment of severe hypertriglyceridemia (SHTG). The drug is designed to specifically target and inhibit the expression of Angiopoietin-like protein 4 (ANGPTL4) mRNA in the liver. ANGPTL4 is a critical regulator of lipid metabolism that acts as an inhibitor of lipoprotein lipase (LPL), the enzyme responsible for the hydrolysis of triglycerides in the bloodstream. By reducing the production of ANGPTL4, A24110He enhances LPL activity, thereby promoting the clearance of triglyceride-rich lipoproteins and significantly lowering plasma triglyceride levels. The GalNAc-conjugation technology facilitates targeted delivery to hepatocytes, improving the drug's potency and safety profile. A24110He is currently being evaluated in clinical trials to address the unmet medical need in patients with extreme elevations in triglycerides who are at high risk for acute pancreatitis and cardiovascular complications.
