A244-rgp120 is a recombinant HIV-1 envelope glycoprotein 120 (rgp120) derived from a Clade E (CRF01_AE) strain, originally identified in Thailand. It was developed as a key component of the AIDSVAX B/E vaccine, which was used in the landmark RV144 Phase 3 clinical trial. Research led by Phil Berman at the University of California, Santa Cruz, has focused on optimizing the production of A244 using CRISPR-engineered MGAT CHO cell lines to ensure the presence of high-mannose glycans, which are essential for the binding of broadly neutralizing antibodies (bNAbs). The protein is currently being evaluated in various prime-boost regimens, including trials like RV306, often in combination with novel adjuvants such as ALFQ or other vaccine candidates like IHV01, to enhance the breadth and durability of the immune response against HIV-1.
