Overview

**A244_N332-rgp120** is a recombinant glycoprotein 120 (rgp120) envelope protein variant derived from the HIV-1 CRF01_AE A244 isolate, engineered with an N-linked glycosylation site shifted from N334 to N332 to enrich high-mannose glycans (primarily Man5). Produced in MGAT1- CHO cells to limit complex glycosylation and sialic acid content, it exhibits improved binding to broadly neutralizing monoclonal antibodies (bN-mAbs) such as PG9 (V1V2 apex), PGT128 and PGT126 (N301/N332 glycans), and 10-1074 (N332-restricted) compared to standard CHO-produced A244-rgp120 used in the RV144 trial. Yields exceed 1 g/L in stable cell lines like 5F MGAT1-CHO, addressing production bottlenecks for HIV vaccine antigens. It aims to enhance immunogenicity over prior rgp120 vaccines by presenting glycan-dependent epitopes absent in complex-glycan versions.
