A2A-O-004 is a small molecule inhibitor of Transforming Acidic Coiled-Coil Protein 3 (TACC3), currently in the IND-enabling phase of development for the treatment of solid tumors. Developed by A2A Pharma using their proprietary SCULPT computational design platform, the drug is engineered to disrupt the interaction between TACC3 and its mitotic partners, such as clathrin. TACC3 is a centrosomal protein essential for the stabilization of kinetochore microtubules during mitosis, and its dysregulation is a hallmark of various aggressive cancers. By targeting this mechanism, A2A-O-004 aims to induce mitotic catastrophe and apoptosis in cancer cells while maintaining selectivity to minimize off-target effects.
