A2aR.KD.MSLN-CAR T cells are genetically engineered chimeric antigen receptor (CAR) T cells designed to target and kill mesothelin-expressing tumor cells in solid tumors, with enhanced resistance to immunosuppression in the tumor microenvironment. These cells co-express an anti-mesothelin CAR and an shRNA that knocks down adenosine 2a receptor (A2aR), reducing the inhibitory effects mediated by adenosine signaling. The A2aR knockdown boosts proliferative, cytotoxic, and cytokine-producing capacities of the CAR T cells even under highly suppressive conditions, making this approach promising for solid tumor immunotherapy[1].
