A2B356 is an investigational autologous CAR T-cell therapy being developed by A2 Biotherapeutics for the treatment of hematological cancers. It utilizes the company's proprietary Tmod (T-cell receptor modulator) platform, which features a dual-receptor design: an activator receptor that targets a tumor-specific antigen and a blocker receptor that recognizes a protective antigen (typically HLA-A*02) on normal cells. This "AND-NOT" logic gate is designed to selectively kill cancer cells that have lost the protective antigen due to loss of heterozygosity (LOH) while sparing healthy tissues. As of early 2024, the specific activator target for A2B356 remains undisclosed, and the program is in the discovery stage.
