A2B530 is an autologous, logic-gated chimeric antigen receptor (CAR) T-cell therapy developed using the proprietary Tmod platform by A2 Biotherapeutics. It is designed to selectively target and kill tumor cells while sparing normal tissues. The therapy incorporates a dual-receptor system: - An activator CAR that targets carcinoembryonic antigen (CEA), which is highly expressed in several solid tumors including colorectal, pancreatic, and non-small cell lung cancers. - A blocker receptor that targets HLA-A*02, a molecule typically lost in tumor cells due to loss of heterozygosity (LOH) but retained on normal cells. This design enables the engineered T cells to attack only those cancer cells expressing CEA and lacking HLA-A*02 expression, thereby reducing on-target/off-tumor toxicity. A2B530 has received FDA Orphan Drug Designation for colorectal cancer and is being evaluated in phase 1/2 clinical trials for colorectal cancer, pancreatic cancer, non-small cell lung cancer (NSCLC), and other CEA-expressing solid tumors with HLA-A*02 LOH[1][3][4][5][7][9][10].
