Overview

A2B694 is an investigational autologous chimeric antigen receptor T-cell (CAR-T) therapy developed by A2 Biotherapeutics. It utilizes the proprietary logic-gated Tmod platform, which incorporates a dual-receptor design to achieve selective killing of tumor cells while sparing normal tissue. The therapy features two CARs on each cell: an activator that targets mesothelin (MSLN), a protein overexpressed in several solid tumors, and a blocker that targets HLA-A*02. In patients whose tumors have lost HLA-A*02 expression (loss of heterozygosity), the blocker prevents activity against normal cells retaining HLA-A*02, thereby reducing on-target off-tumor toxicity. This approach aims to address the challenge of safely targeting solid tumors with CAR-T therapies. Clinical development is focused on MSLN-expressing solid tumors with HLA-A*02 loss such as ovarian cancer, mesothelioma, pancreatic cancer, colorectal cancer, and non-small cell lung cancer[1][3][6][8].
