A3AR siRNA is a small interfering RNA (siRNA) designed to silence the expression of the Adenosine A3 Receptor (A3AR/ADORA3). In biomedical research, particularly in studies of ulcerative colitis (UC), A3AR siRNA is utilized as a tool to investigate the receptor's role in regulating intestinal epithelial cell pyroptosis. Experimental evidence indicates that silencing A3AR increases the activation of Gasdermin D (GSDMD), NLRP3, and caspase-1, thereby promoting the release of pro-inflammatory cytokines like IL-1β and IL-18. This suggests that A3AR signaling normally serves to inhibit GSDMD-mediated pyroptosis, highlighting the receptor as a potential therapeutic target for inflammatory bowel diseases.
