Overview

A49 is a therapeutic peptide developed by Assistance Publique - Hôpitaux de Paris (AP-HP) for the treatment of glioblastoma multiforme (GBM). It is a 12-mer peptide designed to target the **CD44** receptor, a cell-surface glycoprotein that is highly expressed on glioblastoma cells and cancer stem cells. CD44 is a primary receptor for hyaluronic acid (HA), and its activation promotes tumor cell survival, proliferation, migration, and resistance to standard therapies. A49 functions as a CD44 antagonist by binding to the hyaluronic acid-binding domain (HABD) of the receptor, thereby blocking the interaction between CD44 and HA. This disruption inhibits the downstream signaling pathways that drive tumor invasiveness and chemoresistance. A49 is currently being investigated in Phase I/II clinical trials for patients with recurrent glioblastoma.
