Overview

A52-Mel is a melanin-based peptide vaccine developed by Assistance Publique - Hôpitaux de Paris for the treatment of newly-diagnosed glioblastoma. The vaccine targets two specific glioma-associated antigens: telomerase reverse transcriptase (TERT) and protein tyrosine phosphatase receptor type Z1 (PTPRZ1). It is formulated with synthetic melanin and a Toll-like receptor 9 (TLR9) agonist (CpG-ODN) as adjuvants to enhance the immune response. Currently being evaluated in the Phase 1/2a NAVIG-1 clinical trial, the vaccine is administered subcutaneously following standard-of-care radio-chemotherapy and in combination with adjuvant temozolomide. This biologic immunotherapy is designed to stimulate the patient's immune system to recognize and attack glioblastoma cells expressing these tumor-associated antigens, potentially preventing recurrence.
