Overview

A52 peptide is a therapeutic peptide developed by Assistance Publique - Hôpitaux de Paris (AP-HP) and Inserm for the treatment of glioblastoma. It targets Annexin A2 (ANXA2), a calcium-dependent phospholipid-binding protein that is highly expressed in glioblastoma and plays a critical role in tumor cell invasion, migration, and the maintenance of glioblastoma stem cells. The A52 peptide is derived from the sequence of Annexin A2 and acts as a competitive inhibitor, disrupting the interaction of ANXA2 with its ligands, such as S100A10, and preventing the formation of the ANXA2-S100A10 heterotetramer. This disruption inhibits the proteolytic activity and signaling pathways required for tumor progression and invasion into healthy brain tissue. The drug is currently being evaluated in early-phase clinical trials to assess its safety and efficacy in patients with recurrent or newly diagnosed glioblastoma.
