Overview

A6-11-26 is an anti-neuropilin-1 (NRP-1) monoclonal antibody designed as a targeted therapy for hepatocellular carcinoma (HCC). The antibody works by binding to NRP-1, which acts as a co-receptor for vascular endothelial growth factor (VEGF). By blocking NRP-1 interactions with VEGF, A6-11-26 interferes with VEGF signaling pathways that promote tumor angiogenesis, growth, and metastasis. The mechanism involves preventing formation of the VEGF-NRP-1-VEGFR ternary complex, which is critical for receptor internalization and activation of downstream intracellular signaling pathways. Additionally, A6-11-26 may inhibit c-Met, reduce p130Cas phosphorylation, and modulate the tumor microenvironment. The antibody demonstrates dose-dependent anti-proliferative effects that correlate with NRP-1 expression levels in tumor cells, showing strongest efficacy in HCC with high NRP-1 expression. A6-11-26 has been shown in preclinical studies to inhibit tumor cell proliferation, reduce colony formation, promote apoptosis, and decrease expression of both NRP-1 and VEGFR in tumor tissues.
